Finally a third church dedicated to Our Lady of the Most Holy Rosary was built in 1596 where the current Binondo Church stands. This third church is the sole survivor of those early structures after numerous reconstructions.

The chapel’s significance is deeply rooted in the history of evangelization within the Chinese community in Manila. Early missionaries including Fr Miguel de Benavides later Archbishop of Manila and founder of the University of Santo Tomas played a key role in converting Chinese merchants. The Dominican Fathers played a crucial role in ministering to the Chinese community.

The path to the present chapel was not always smooth. World War II brought destruction. Bombings in 1944 razed the Binondo Church. A small church was later built on the site of the old convent. It was inaugurated in 1945. The current image of Our Lady of the Holy Rosary was inaugurated and blessed on Easter Sunday 1947. This image became the focal point of devotion for the Filipino-Chinese community. A replica was later commissioned for processions.

Today Our Lady of the Most Holy Rosary Chapel continues to thrive. It serves as a place of worship and a symbol of cultural and religious harmony.
