Albert Levame became the first Apostolic Nuncio on November 12, 1939. He served until October 3, 1949. Other notable Nuncios include Alfredo Pacini and Raffaele Forni. They each played a role in shaping the Nunciature.

Andrea Cordero Lanza di Montezemolo served as Nuncio from 1986 to 1990. Janusz Bolonek held the position from 1999 to 2008. More recently, Martin Krebs served until March 3, 2021. Gianfranco Gallone is the current Apostolic Nuncio. He assumed the role on January 3, 2023.

Popes have also directly engaged with Uruguay. Pope John Paul II made two pastoral visits. These visits occurred in 1987 and 1988. In June 2013, President Josè Mujica visited Pope Francis. These events highlight the ongoing dialogue.
