The Holy Trinity Cathedral was built between 1860 and 1872. It was constructed by the Russian Orthodox Ecclesiastical Mission. At that time, Jerusalem was part of the Ottoman Empire. The increased flow of Christian pilgrims to the Holy Land in the mid-19th century prompted its construction. Tsar Alexander II helped the Russian Church build hostels and services for these pilgrims. This area became known as the Russian Compound.

Martin Eppinger, a German-Russian architect, designed the Holy Trinity Cathedral. Initially, it was to be dedicated to St Alexander Nevsky. He was the patron saint of Alexander II. Ultimately, it was decided to dedicate it to the Holy Trinity.

The Holy Trinity Cathedral has a cruciform layout, a basilica with a central nave and two side aisles. Two lateral rooms in rounded apses complete the cross shape. Built from white stone, the cathedral features eight steeples. Each steeple is topped with a cross. Inside, murals cover the walls. True to Orthodox tradition, the cathedral has few chairs. Worshippers typically stand during services.

The Cathedral of the Holy Trinity was officially founded on September 11, 1860. By 1864, the exterior was complete. Construction was then suspended due to budget issues. In 1872, the cathedral was finally inaugurated. Grand Duke Nikolai, brother of Alexander II, attended the ceremony. Since then, pilgrims and locals have used the Holy Trinity Cathedral.

Renovations took place from 1895 to 1897. Workers repaired the roof and fixed the blind area. They scraped off glutinous paints from the walls. Three interiors were painted white. Then, sacred images were added to decorate the cathedral. An emergency exit was built next to the altar.

During the British Mandate, the Trinity Cathedral remained independent. It fell under the jurisdiction of the Russian Orthodox Church Outside of Russia. The first Arab-Israeli war caused damage to the church. Since 1948, it has been under the Moscow Patriarchate of the Russian Orthodox Church. Ownership of the church remains undetermined. This is due to an ongoing legal and diplomatic dispute between Russia and Israel.

The Holy Trinity Cathedral is not large. It was originally intended for pilgrims staying in the Russian Compound. It was assumed they would prefer the historic churches in the Old City. However, its height and magnificent style make it a prominent structure.
