The foundation stone was laid in 1962. The Hamburg Building Authority designed the original school buildings. These included classrooms, specialist rooms, connecting corridors, and a gymnasium. The first phase of construction was inaugurated in 1965. The gymnasium was added in 1970. Administration followed in 1971. School operations moved to the new building in 1965. It moved from Sinstorfer Kirchweg.

In 2010, as part of Hamburg’s school reforms, the Haupt- und Realschule Sinstorf merged. It merged with the Haupt- und Realschule Hanhoopsfeld and the Lessing-Aufbaugymnasium. The new school was initially called Stadtteilschule Hanhoopsfeld/Sinstorf. It was later renamed Lessing-Stadtteilschule.

In 2019, the Lessing-Stadtteilschule was consolidated. It was in new buildings at the Hanhoopsfeld site. The Sinstorfer Weg school was vacated. Since the 2020/21 school year, the newly founded Grundschule Sinstorfer Weg has used the site. Buildings on the south of the school grounds were demolished by 2021. A new building is planned.
